Analysis of the diagnostic prestatien of the industrial RT-PCR kits for the detection of SARS-CoV-2 in Colombia

The ongoing SARS-CoV-2 pandemic has led to the design and development of several RT-PCR kits designed to enable rapid scale-up of molecular exams for massive screening. We evaluated the diagnostic performance of nine commercial kits that showed optimal performance and high discriminatory power. However, we observed differences in sensitivity, specificity and E-Gen-Ct values ​​and discussed these results in light of the influence of  the  genetic variability  of SARS-CoV-2  and its possible impact on current molecular diagnostic assays. Antimikrobielle Aktivität von Kamferboomzilvernanodeeltjes tegen vuilbroedziekten en het ontdekken van een nieuwe stam van  Serratia marcescens mittels DGGE-PCR  als Sekundärinfektion auf Honingbijlarven verabreicht wurde

Amerikanische Faulbrut (AFB) und Europäische Faulbrut (EFB) sind die beiden wichtigsten bakteriellen Krankheiten, von denen Honigbienen betroffen sind. Dies führt zu einer Verringerung der Lebensfähigkeit des Bienenstocks, einer Verringerung der Honigproduktion und zu erheblichen wirtschaftlichen Verlusten für die Imker.

Aufgrund der Ineffizienz und / oder geringen Wirksamkeit einiger Antibiotika stellen Forschungen mit Nanotechnologie möglicherweise neue therapeutische Strategien dar. Nanostruktur-Medikamente haben gegenüber herkömmlichen Medikamenten einige Vorteile gezeigt , wie langsame, schrittweise und kontrollierte Freisetzung, erhöhte Bioverfügbarkeit und verringerte Nebenwirkungen.

In dieser Studie wurden verschiedene infizierte Larven aus zwei Bienenhäusern gesammelt; Die Kämme, die Symptome einer amerikanischen und europäischen Faulbrut aufwiesen, wurden isoliert.

Die  antimikrobielle In-vitro- Aktivität von Kampferbaum-Silber-Nanopartikeln gegen Faulbrutkrankheiten wurde unter Verwendung von UV-Vis-Spektrophotometrie und Rasterelektronenmikroskop (SEM) charakterisiert , was die Bildung von Silbernanopartikeln mit einem Größenbereich von 160 bis 660 nm belegt. Die antimikrobielle Aktivität der Silbernanopartikel wurde unter Verwendung eines Agardiffusionsassays getestet und bewies ihre Fähigkeit, das pathogene Bakterienwachstum sowohl in AFB als auch in EFB wirksam zu stoppen .

Die DGGE-PCR-Technik wurde zur Identifizierung seltener bakterieller Infektionen von Honigbienen in Abhängigkeit von der 16S-rRNA-Amplifikation aus ihrer gesamten extrahierten DNA angewendet und als Serratia marcescens  (TES) identifiziert  , die in der GenBank mit einer neuen Zugangsnummer (MT240613) hinterlegt ist.

Die Ergebnisse bestätigten, dass der Stamm durch DGGE-PCR-Analyse nachgewiesen wurde, die eine einzigartig infizierte Brut verursachte, die von der amerikanischen Foulbrood angegriffen wurde. Es konnte gefolgert werden, dass grün synthetisierte Silbernanopartikel voraussichtlich als wirksame Behandlung für bakterielle Honigbienenkrankheiten eingesetzt werden. Diese Materialien müssen unter Feldbedingungen genauer untersucht werden und untersuchen die Möglichkeit ihrer Rückstände in Honigbienen wie Honig und Bienenwachs.
